Generation of Leukotrienes by Purified Human Lung
نویسنده
چکیده
separation to purify human lung mast cells to 98% homogeneity. Lung cell suspensions were obtained by dispersion of chopped lung tissue with proteolytic enzymes. Mast cells were then purified from the suspensions by countercurrent centrifugal elutriation and affinity chromatography. The purified mast cells released both histamine and slow-reacting substance of anaphylaxis (SRS-A) (leukotriene C and D) during stimulation with goat anti-human IgE antibody. Moreover, these preparations were able to generate significant quantities of SRS-A (32±7X 10-'7 LTD moleequivalents/mast cell) at all stages of purification, indicating that a secondary cell is not necessary for the antigen-induced release of SRS.

Lipoxygenase(LOX) catalyzes irreversible transfer of oxygen molecule to Arachidonic and Linoleic acid to produce 13 Hydroproxy Octadecadienoic acid. Recent studies showed the involvement of Lipoxygenase products, leukotrienes, in inflammations and Lipoxygenase pathways acts as mediators of early inflammatory events in atherosclerosis. The aim of the present study was purification and characteri...
متن کاملTHE PRODUCTION OF MURINE MONOCLONAL ANTIBODIES (MAb) DIRECTED AGAINST HUMAN T- LYMPHOCYTE SUBSETS
The production of murine monoclonal antibody (MAb) has not yet been reported in Iran. The present work describes for the first time the generation of several murine hybridoma clones secreting MAbs directed against human leukocyte surface antigens. The secreted antibodies by hybridoma clones have been screened on different lymphoid and non-lymphoid tissues. Results indicated that of seven h...
متن کاملTwo-dimensional Simulation of Mass Transfer and Nano-Particle Deposition of Cigarette Smoke in a Human Airway
The chance of developing lung cancer is increased through being exposed to cigarette smoke illustrated by studies. It is vital to understand the development of particular histologic-type cancers regarding the deposition of carcinogenic particles, which are present in human airway. In this paper, the mass transfer and deposition of cigarette smoke, inside the human airway, are investigated apply...
متن کاملBiologic activity of purified cotton bract extracts in man and guinea pig.
Purified aqueous extracts of cotton bract induce acute airway constriction in healthy volunteers never before exposed to cotton bract. The response is similar to that of textile workers who inhale cotton dust. Approximately 60% of volunteers respond to bract extract with significant decreases in lung function, and these volunteers show an increased number of lymphocytes present in their lungs. ...
متن کاملEpoxyeicosatrienoates are the dominant eicosanoids in human lungs upon microbial challenge
Lipoxygenase, cyclo-oxygenase and cytochrome P450 (CYP) products of arachidonic acid (AA) are implicated in pulmonary vasoregulation. The CYP-mediated epoxyeicosatrienoates (EETs) have been described previously as the predominant eicosanoids in human lungs upon stimulation with the Ca ionophore A23187. In this study, we challenged perfused human lungs with two microbial agents: Escherichia coli...
